Research Interests: Arteries supply critical nutrients to metabolically active tissues of the body and have an intricate structure which includes a single inner cell layer of endothelial cells (ECs) surrounded by multi-layered vascular smooth muscle cells (VSMCs). A variety of vascular diseases, such as pulmonary artery hypertension and coronary artery atherosclerosis, are characterized by excessive and disordered EC and VSMC proliferation and migration. Many of the molecular mechanisms underlying these findings are likely to recapitulate the processes regulating morphogenesis of developing vessels. However, the development of the VSMC and EC layers of vessels is a poorly understood process. We are studying the morphogenesis of the left pulmonary artery of the embryonic mouse and our work, to date, has made this vessel one of the best described process of artery formation in vivo. Consequently, this work promises to have important implications for both vascular biology and clinical disorders characterized by aberrant EC and VMSC physiology.

Research Interests: #1. Thrombin biochemistry and biology - we have generated a large panel of thrombin mutants by alanine scanning site-directed mutagenesis, and used them to characterize thrombin's interaction with various substrates. We are interested in the physiological mechanisms that regulate thrombin's prothrombotic and pro-inflammatory functions and in the development of novel antithrombotic drugs. #2. Novel endothelial cell markers - we have identified a number of novel endothelial cell transcripts using SAGE and microarray transcript profiling. The biochemistry and cell biology of some of these novel endothelial proteins are being pursued.

Research Interests: Physiological underpinnings of cognitive impairment in older adults. In particular, I am interested in the contribution of cardiovascular disease to cognitive decline and/or the onset of Alzheimer's disease and vascular dementia. Additionally, I have conducted studies and published on the impact of Coronary Artery Bypass Surgery on cognition in the elderly. A central part of my work is on the impact of the genetic risk factor for dementia, Apolipoprotein E4 allele, on cognition and its involvement in mediating the relationship between cardiovascular disease and dementia.
