Thrombin biochemistry and biology - we have generated a large panel of thrombin mutants by alanine scanning site-directed mutagenesis, and used them to characterize thrombin's interaction with various substrates. We are interested in the physiological mechanisms that regulate thrombin's prothrombotic and pro-inflammatory functions and in the development of novel antithrombotic drugs. #2. Novel endothelial cell markers - we have identified a number of novel endothelial cell transcripts using SAGE and microarray transcript profiling. The biochemistry and cell biology of some of these novel endothelial proteins are being pursued.

In particular, I am interested in the contribution of cardiovascular disease to cognitive decline and/or the onset of Alzheimer's disease and vascular dementia. Additionally, I have conducted studies and published on the impact of Coronary Artery Bypass Surgery on cognition in the elderly. A central part of my work is on the impact of the genetic risk factor for dementia, Apolipoprotein E4 allele, on cognition and its involvement in mediating the relationship between cardiovascular disease and dementia.
